Introduction

One of the most commonly prescribed drugs for ED, sildenafil citrate (Viagra) is a phosphodiesterase inhibitor that works by increasing blood flow to the penis. It is available in tablet form as well as in an oral solution. In this study, researchers evaluated the efficacy and tolerability of sildenafil citrate 100 mg once daily for 12 weeks in patients with ED who had not previously used sildenafil.

Erection hardness significantly improved for 74% of men taking sildenafil citrate for the first time, according to data from an open-label extension study.

The first open-label extension study of sildenafil citrate 100 mg, published in this issue (May 28) by Vial et al., is a large, multicenter trial that compared the effectiveness of sildenafil citrate to placebo and found that it was effective in improving erection hardness for 74% of men taking the drug for the first time.

The results revealed that 26% of men had improved intercourse success after taking sildenafil citrate compared with those who received a placebo treatment.

Intercourse success increased from 26% to 66%.

The percentage of patients who reported intercourse success increased from 26% to 66%. The percentage of patients who reported intercourse success for the first time increased from 74% to 92%.

The most common side effect was headache, which occurred in 19% of men treated with the 100 mg dose.

Headache is a common side effect of sildenafil citrate and can be treated with over-the-counter medications. It may also be treated with a lower dose of sildenafil citrate, but this would require more frequent dosing to maintain an erection.

In addition to improvement in erection hardness, investigators assessed erection quality using the Erectile Dysfunction Inventory of Treatment Satisfaction (EDITS) questionnaire.

In addition to improvement in erection hardness, investigators assessed erection quality using the Erectile Dysfunction Inventory of Treatment Satisfaction (EDITS) questionnaire. The EDITS is a validated questionnaire that explores the patient's overall satisfaction with his erections as well as their ability to have intercourse after oral therapy.

A greater percentage of patients completing 4 weeks' treatment experienced improved erection quality (30%) compared with earlier in the study (17%).

Erection hardness was significantly better in those treated for 4 weeks than at baseline or after 1 week of treatment.

This extension study demonstrated that one dose of sildenafil citrate 100 mg provided consistent and statistically significant improvement in erection hardness and intercourse success over 12 weeks.

This extension study demonstrated that one dose of sildenafil citrate 100 mg provided consistent and statistically significant improvement in erection hardness and intercourse success over 12 weeks.

A total of 854 patients were randomized to receive either placebo or sildenafil citrate 100 mg, once daily for 12 weeks. The primary endpoint was the mean change from baseline in erectile function (EF) domain score from week 0 to week 12. Secondary endpoints were EF domain score at end of treatment (week 10), percent change from baseline at end of treatment (week 10), changes in ESS score between baseline and week 10, and QOL questionnaires during the first 3 days after starting therapy with placebo or sildenafil citrate 100 mg

Conclusion

The results of this study indicate that sildenafil citrate 100 mg may be a useful starting point for patients who have not used sildenafil before and are uncertain about their response to it. The results also demonstrate that an extension study is necessary to confirm the effectiveness of long-term use over time.
